While saving the app-v 5.1 package on win 10 virtual machine, i am getting below error. Anyone have seen this error before?
failed to load virtual services information. - failed to create a manifest instance

Are you using the App-V 5.1 sequencer? Could try the the newer sequencer from the ADK, it has all the bug fixes etc.
Another thing to check, though its a long shot, export your manifest and search for "~" (tilde symbol*). I've seen the past the sequencer malforming the App-V directory tokens.
*not sure if you can see the tilde, looks like a dash in my comment //shrugs.
